no.
you don't need reinstall operating system or of applications.
you need restart server 3 times. below detailed how-to in hig level steps:

  1. shut down server , insert second disk
  2. open computer management (compmgmt.msc) , go disk management
  3. in right pane select disks. convert them dynamic disks.
  4. reboot twice, when asked.
  5. go disk management , select 1 of partitions. right click , select 'add mirror'.
choose make mirror on second hard disk.
  6. select other partition want mirror. right click , select 'add mirror' disk too.
choose create the mirror on second hard disk well.

now, when restart server see second boot option, called secondary plex. when first hard disk fails able boot second hard disk.

note:
disk mirroring provides fault tolerance @ best. not protect against disk corruption. when data on 1 of disks gets corrupted, both disks corrupted. when rogue update or driver makes first drive unbootable, second drive unbootable. protect against these kinds of circumstances, implement backup practice.
